Clinical Report: Correction to PAK4 Inhibition and PD-1 Blockade

Overview

A correction published online on 9 December 2019 clarifies how the mouse data in Fig. 5a of the Nature Cancer article were assembled. The n = 16 mice shown per group were pooled across three independent experiments; the legend and source data file were amended, and the correction was incorporated into the HTML and PDF versions.

Background

The corrected article concerns PAK4 inhibition in combination with PD-1 blockade immunotherapy. Figure 5a presented mouse-group data pooled across independent experiments, while Fig. 5d showed one experiment within that set. Clarifying the experiment-level sample sizes and pooling is relevant to interpreting the figure and its associated source data. The correction addresses reporting details and does not provide new efficacy results.

Data Highlights

Figure or groupSample size and experiment details
Fig. 5an = 16 mice per group, pooled across three independent experiments
Fig. 5d: PAK4 KO isotypen = 6 mice in one experiment
Fig. 5d: PAK4 KO anti-PD1n = 6 mice in one experiment
Fig. 5d: PAK4 KO anti-PD1 anti-CD8n = 5 mice in one experiment

Key Findings

  • The correction was published online on 9 December 2019 and concerns the Nature Cancer article with DOI 10.1038/s43018-019-0003-0.
  • The original Fig. 5 legend and corresponding source data file did not state that the n = 16 mice per group in Fig. 5a were pooled across three independent experiments.
  • Fig. 5d showed one of those experiments, with n = 6 mice each in the PAK4 KO isotype and PAK4 KO anti-PD1 groups.
  • The same Fig. 5d experiment included a PAK4 KO anti-PD1 anti-CD8 group with n = 5 mice.
  • The Fig. 5 legend was amended, and the pertinent experiments were identified in the associated source data file.
  • The correction was incorporated into both the HTML and PDF article versions.

Clinical Implications

This correction clarifies the sample-size pooling and experiment-level group sizes needed to interpret Fig. 5 and its source data. It does not report new clinical evidence or alter the article’s findings.

Conclusion

The correction documents the pooled nature of the Fig. 5a mouse data and specifies the group sizes in the experiment shown in Fig. 5d. The article’s online and PDF versions were updated accordingly.
