To evaluate the feasibility of patient-derived tumor organoids (PDTOs) in predicting treatment responses for patients with advanced solid tumors.
Approach:
Sample Analysis: Retrospective analysis of 164 solid tumors for organoid establishment and a clinical cohort of 23 patients to validate PDTOs' predictive performance.
Drug Sensitivity Testing: PDTO-based drug sensitivity testing was conducted to assess concordance with clinical treatment outcomes.
Key Findings:
The overall establishment success rate of organoids reached 91.5%, with rates ranging from 90.6% to 92.6% across different cancer types.
Histopathological analysis confirmed that PDTOs partially recapitulated the histopathological features and proliferative activity of matched original tumors.
PDTO-based drug sensitivity testing showed good concordance with clinical treatment outcomes, yielding an overall accuracy of 85.0%, a sensitivity of 86.7%, and a specificity of 80.0%.
Interpretation:
PDTOs may serve as a companion diagnostic tool for personalized cancer therapy and assist in evaluating multidrug resistance in advanced solid tumors.
