A one-pot method detects and characterizes phospholamban proteoforms from biopsy-scale samples
Objective:
To develop a simplified extraction method for analyzing phospholamban using reduced cardiac tissue.
Approach:
- Extraction Method: A one-pot method using Azo surfactant was developed to extract phospholamban from 1 mg of cardiac tissue.
- Testing: The method was tested on left ventricular tissue from a nonfailing adult donor heart, using sonication to enhance protein extraction.
- Analysis: Intact proteins were analyzed using liquid chromatography-tandem mass spectrometry to distinguish proteoforms.
Key Findings:
- The new method reduced the required tissue from 10 mg to 1 mg while maintaining similar analytical performance.
- The method produced 30 to 60 µg of total protein from 1 mg of cardiac tissue.
- Phosphorylation measurements were comparable to those obtained using the conventional method.
Limitations:
- The method was evaluated using tissue from only one nonfailing donor.
- It focused solely on phospholamban and did not assess the full cardiac proteome.
- The palmitoylated form of phospholamban was not confirmed by targeted tandem mass spectrometry.
Conclusion:
Further research is needed to validate the method in clinical biopsy samples and diseased tissues.
