To examine how diet-induced obesity affects haematopoietic responses in the bone marrow and spleen, along with myeloid cell infiltration after myocardial infarction (MI).
Approach:
Mouse Model: Male and female C57BL6/J mice were fed either a high-fat or low-fat diet for 16 weeks before inducing MI through permanent ligation of the left anterior descending coronary artery.
TREM1 Blockade: The therapeutic effect of TREM1 blockade was assessed using the LR12 peptide administered post-MI, with control animals receiving a scrambled peptide.
Echocardiography: Echocardiography was performed on days 5 and/or 56 post-MI to evaluate cardiac function.
Key Findings:
Obese mice exhibited enhanced myelopoiesis after MI, particularly in the spleen.
Infarcted myocardium from obese mice showed increased expression of TREM1.
Serum from obese mice post-MI increased TREM1 expression on bone marrow cells.
Limitations:
Study conducted in a mouse model, which may not fully replicate human conditions.
Focus on short-term outcomes post-MI without long-term follow-up.
The proposed framework classified 30% of patients as high or extreme risk, compared with 12% classified as having severe or greater tricuspid regurgitation under established grading schemes.
