Objective:

To examine how diet-induced obesity affects haematopoietic responses in the bone marrow and spleen, along with myeloid cell infiltration after myocardial infarction (MI).

Approach:
  • Mouse Model: Male and female C57BL6/J mice were fed either a high-fat or low-fat diet for 16 weeks before inducing MI through permanent ligation of the left anterior descending coronary artery.
  • TREM1 Blockade: The therapeutic effect of TREM1 blockade was assessed using the LR12 peptide administered post-MI, with control animals receiving a scrambled peptide.
  • Echocardiography: Echocardiography was performed on days 5 and/or 56 post-MI to evaluate cardiac function.
Key Findings:
  • Obese mice exhibited enhanced myelopoiesis after MI, particularly in the spleen.
  • Infarcted myocardium from obese mice showed increased expression of TREM1.
  • Serum from obese mice post-MI increased TREM1 expression on bone marrow cells.
Limitations:
  • Study conducted in a mouse model, which may not fully replicate human conditions.
  • Focus on short-term outcomes post-MI without long-term follow-up.
