To highlight recently published research by Dana-Farber faculty from June 1 - 15, 2026.

Approach:
  • COSMIC-313 Study: Phase III study comparing cabozantinib plus nivolumab and ipilimumab versus placebo plus nivolumab and ipilimumab in advanced renal cell carcinoma.
  • SOFT and TEXT Trials: Combined analysis of trials evaluating the impact of ovarian function suppression and endocrine therapy on premenopausal women with hormone receptor-positive early breast cancer.
  • NSD2 in Multiple Myeloma: Investigation of transcriptional targets dysregulated by NSD2 in t(4;14) multiple myeloma.
  • PKMYT1 in Multiple Myeloma: Exploration of PKMYT1 as a targetable vulnerability in del(17p) high-risk multiple myeloma.
Key Findings:
  • Cabozantinib plus nivolumab and ipilimumab showed longer progression-free survival (16.6 months) compared to placebo (11.2 months) in advanced renal cell carcinoma.
  • No significant difference in overall survival was observed between treatment arms in the COSMIC-313 study.
  • In the SOFT trial, 15-year disease-free survival was 78.6% for exemestane plus ovarian function suppression, compared to 72.1% for tamoxifen alone.
  • High-risk premenopausal subgroups showed meaningful overall survival benefits from adjuvant exemestane and/or ovarian function suppression.
  • NSD2 overexpression in multiple myeloma is linked to the t(4;14) translocation and affects transcriptional regulation.
Interpretation:

Limitations:
  • The COSMIC-313 study did not show overall survival benefits despite progression-free survival advantages.
  • The SOFT and TEXT trials' findings may not be generalizable to all premenopausal women with breast cancer.
