Objective:

To report multicenter outcomes of CD3xCD20 bispecific antibodies (BsAbs) in patients with active or prior history of CNS involvement by aggressive B-cell non-Hodgkin lymphoma (B-NHL).

Key Findings:
  • In patients with active CNS involvement (n=18), the overall response rate (ORR) was 56%, with a complete response (CR) rate of 33%.
  • In patients with prior CNS involvement (n=10), the ORR was 60%, with a CR rate of 50%.
  • The median time to best CNS response was 58 days.
  • 6-month progression-free survival (PFS) was 43% for active CNS involvement and 46% for prior CNS involvement.
  • 6-month overall survival (OS) was 77% for active CNS involvement and 89% for prior CNS involvement.
Interpretation:

Limitations:
  • The study is retrospective and includes a small sample size.
  • Patients with active CNS involvement were excluded from pivotal trials, limiting generalizability.
  • The safety profile in patients with prior CNS involvement but no active disease is not fully characterized.
