Whole-transcriptome sequencing separated 24 CEBP-altered B-ALL cases into two distinct transcriptomic clusters across pediatric, AYA, and adult patients.

  • 2

Cluster A aligned with CEBP/ZEB2 and was enriched for ZEB2 p.H1038R, NRAS mutations, and CDKN2A deletions.

  • 3

Cluster B aligned with CEBPalt and featured FLT3 mutations, frequent IKZF1 deletions, and a higher overall burden of karyotypic gains and losses.

  • 4

CEBPE rearrangements occurred only in Cluster A, whereas CEBPD rearrangements occurred exclusively in Cluster B.

  • 5

The clusters showed different B-cell developmental signatures, with pre-B I enrichment in Cluster B and pre-B II Small or Immature B-cell enrichment in Cluster A.
